Nikon Ti-HCS microscope

Profile: High content screening system for multiwell plates in phase contrast and/or fluorescence.

Here we have a fully automated, inverted Nikon Ti microscope made for high content screening tasks. The microscope is encased by a evironmental box from Oko Lab for temperature-, CO2- and humidity control. By a software module names "JOBS" in the Nikon NIS-Elements acquisition software it is possible to screen in x/y, z and t everything from a LabTek dish to a multiwell plate. Images can be taken in brightfield and epifluorescence.

Objectives

  • Nikon Plan Fluor 10x NA 0.3 phase contrast (working distance 16mm; corrected for 0.17mm coverslips (= type #1.5))
  • Nikon S Plan Fluor ELWD 40x NA 0.60 (working distance 3.6 - 2.8mm; adjustable for glas and plastic bottom dishes with a thickness between 0 and 2mm).
  • Nikon Plan Fluor 20x NA 0.75 Multi Immersion (working distance 0.35mm; water, glycerol. oil; adjustable for glas bottom dishes of type #1 - 2)
  • Nikon Plan Apo VC 20x NA 0.75 (working distance 1.0 mm; corrected for 0.17mm coverslips (= type #1.5))

Camera

Andor Clara (model DR-328-C01-SIL) high-sensitive interline-CCD-camera with 1392 x 1040 pixel resolution. Each pixel has a size of 6.45 x 6.45 μm. The pixel well factor is 18.000 electrons. The CCD is cooled down thermoelectrically to -45°C and has a readout rate of 11 frames per second in full resolution.

Fluorescent light source

Lumencor SpectraX light engine. A LCD light source offering the following excitation wavelengths:

  • 395 nm
  • 440 nm
  • 475 nm
  • 508 nm
  • 560 nm
  • 640 nm
